And in laboratories around the world, scientists are now transforming this molecule of heredity into a material of engineering—a hydrogel that is simultaneously rubber-like in its elasticity, steel-like in its toughness, and biological in its compatibility.

The breakthrough, published in Nature Communications in January 2026, represents a fundamental shift in how we think about DNA. Researchers at Huazhong University of Science and Technology have developed a Fast-Shrinking-Induced Entanglement (FaSIE) process that produces hydrogels composed solely of DNA with mechanical properties that rival synthetic elastomers: stiffness exceeding 800 kPa, toughness exceeding 5 MJ/m³, and stretchability exceeding 1000%. These are not the soft, jelly-like DNA hydrogels of the past—they are robust, resilient, and engineering-ready.

The mechanical performance of FaSIE DNA hydrogels is nothing short of extraordinary.

Stiffness > 800 kPa: This places the material in the range of soft structural tissues like cartilage (10–25 kPa) and skin (10–100 kPa), and approaching the lower end of bone (1 MPa). It is orders of magnitude stiffer than conventional DNA hydrogels, which typically have elastic moduli below 50 kPa.

Toughness > 5 MJ/m³: Toughness measures a material's ability to absorb energy before fracture. Five megajoules per cubic metre is comparable to many synthetic elastomers and engineering plastics. It means the material can withstand significant mechanical stress without failing.

Stretchability > 1000%: The hydrogels can be stretched to more than ten times their original length without breaking. This is the hallmark of elastomeric materials—the ability to undergo large reversible deformations. The average elongation at break of these gels reaches 1107%.

These properties are achieved without any chemical crosslinking, synthetic polymer additives, or toxic processing agents. The material is pure DNA, processed through physical dehydration.

Programmable Hydrogels: Tuning Mechanics with DNA Architecture

The FaSIE process is not the only breakthrough in DNA hydrogel engineering. Researchers are also exploiting DNA's inherent programmability to create hydrogels with precisely controlled mechanical properties.

A study published in npj Biomedical Innovations in 2026 developed stiffness-tuneable DNA supramolecular hydrogels using different branching architectures—Double Crossovers (DX), Paranemic Crossovers (PX), and Tensegrity architectures. These hydrogels achieve widely adjustable mechanical properties ranging from 50 to 185 kPa without requiring chemical additives or enzymatic crosslinking.

The significance lies in the ability to programme cellular and organellar states. By tuning the mechanical stiffness of the hydrogel matrix, researchers can influence cellular behaviour in retinal pigment epithelial cells: enhancing cellular adhesion, encouraging elongation (a 3–8-fold increase in area compared to control), and improving viability (1–8-fold increase). This work presents a framework for automating the production of stiffness-tunable DNA hydrogel scaffolds, aligning with the mechanical needs of various cells and tissues and advancing personalised, high-throughput tissue engineering platforms.

Another approach uses sticky-end ligation to freeze network topology and enhance mechanical stability. Ligation extends DNA hydrogel lifetime in serum by at least sevenfold and stiffens the material, enabling facile handling and 3D printing.

Protein-Reinforced DNA Hydrogels

Researchers have also developed a novel one‑pot, two‑step fabrication strategy in which long DNA strands produced by rolling circle amplification form an initial viscoelastic network that is subsequently reinforced through controlled thermal self‑assembly of protein. This synergistic protein‑reinforcement enables precise control over morphological and mechanical properties while improving stability under enzymatic and pH stress. The resulting BRIDGe hydrogels maintain structural integrity under complex deformation and sustained compression in ex vivo nucleus pulposus models and exhibit pressure‑dependent drug release. This establishes protein‑reinforcement within viscoelastic gel networks as a distinct materials design strategy for creating programmable biomaterials that integrate molecular precision with mechanical resilience for use in mechanically demanding biological environments.

Biomedical Applications: Where DNA Meets Medicine

The combination of biocompatibility, programmability, and now mechanical robustness makes DNA hydrogels extraordinarily versatile for biomedical applications. Recent reviews have highlighted their potential in cell culture, drug delivery, tissue engineering, immunotherapy, and biosensing.

Drug Delivery

DNA hydrogels retain a hydrophilic three‑dimensional network structure similar to biological tissues, exhibiting high biocompatibility, programmable responsiveness, and specific recognition functions. The adjustable porosity of these hydrogels allows for controlled release of therapeutic agents, significantly improving efficacy and targeting capabilities.

The FaSIE process enables the production of mechanically robust DNA hydrogels suitable for injectable fillings and localised drug delivery. Protein‑reinforced BRIDGe hydrogels exhibit pressure‑dependent drug release, making them ideal for load‑adaptive therapeutics in mechanically active environments such as the intervertebral disc.

Tissue Engineering

DNA hydrogels are utilised as versatile scaffolds for tissue engineering, providing a three‑dimensional, porous, and hydrated extracellular matrix‑mimicking microenvironment for cell growth and development.

A dual‑functional mineralised DNA hydrogel system (Cap‑gel) integrates immunomodulation and osteogenesis for bone regeneration. The stable and programmable spatial structure of tetrahedral framework nucleic acids promotes macrophage polarisation toward the M2 phenotype while serving as a nucleation template for calcium phosphate crystallisation. In vivo, Cap‑gel modulated early immune responses and accelerated bone regeneration in a calvarial defect model.

Stiffness‑tuneable DNA supramolecular hydrogels with different branching architectures demonstrate excellent strain‑bearing and load‑bearing capacity, making them suitable for biomedical applications. The hydrogels influence cellular behaviour by enhancing cellular adhesion, encouraging elongation, and improving viability.

Cancer Therapy and Immunotherapy

DNA hydrogels are emerging as powerful platforms for cancer therapy. A spatiotemporally controllable DNA hydrogel mesh (SNARE) has been developed for focused antimetastasis therapy. DNA‑based bioinks are advancing biofabrication. An electrochromic DNA‑based bioink with rapid interfacial gelation responds to dynamic biological and electrical cues, making it suitable for emerging applications including wearable devices and biosensors.

3D printing technology enhances nucleic acid‑based molecular diagnostic platforms and wearable biosensors for real‑time monitoring, thereby providing robust support for early disease diagnosis and facilitating the development of targeted and controlled drug delivery systems.

Beyond Medicine: Energy, Robotics, and Sustainability

Energy Storage

DNA hydrogels are being explored as components in batteries, supercapacitors, and fuel cells. Compared to traditional materials, DNA hydrogels offer unique advantages: programmability, biocompatibility, and the ability to form three‑dimensional networks conducive to ion transport. This is an emerging field with significant potential.

Soft Robotics

The FaSIE process enables the fabrication of DNA‑based soft magnetic robots. The combination of mechanical robustness, stretchability, and biocompatibility makes DNA hydrogels ideal for soft robotic applications—devices that must deform, adapt, and interact safely with biological tissues.

Cost: DNA extraction and purification are currently more expensive than synthetic polymer production. However, the abundance of biomass DNA and advances in extraction technology are rapidly reducing costs.
